Imaging-Enabled Orthotopic PDAC Spheroid Models for Longitudinal MRI and Ultrasound Assessment
Using patient-derived 3D spheroids and multimodal imaging to establish and longitudinally monitor a clinically relevant preclinical pancreatic cancer model

Pancreatic ductal adenocarcinoma (PDAC) remains one of the most aggressive and treatment-resistant cancers, creating a need for preclinical models that more closely reflect human tumor biology and disease progression.
In this study, researchers established an orthotopic PDAC model using patient-derived 3D spheroids implanted into NSG mice. Tumor establishment and progression were monitored longitudinally using high-frequency ultrasound and high-resolution anatomical MRI, enabling non-invasive assessment of tumor growth and morphology over time. The model combines biologically relevant 3D tumor systems with advanced imaging to provide a platform for investigating PDAC biology and evaluating potential therapeutic strategies.
