Setting the Standard: Empirical Benchmarks for Case-level Site–Central Discordance in Radiographic Endpoints in Oncology Clinical Trials

Setting the Standard: Empirical Benchmarks for Case-level Site–Central Discordance in Radiographic Endpoints in Oncology Clinical Trials

Contemporary Clinical Trials Communications, September, 2026

Disagreement between site investigators and blinded independent central review (BICR) is a well-recognized reality in oncology clinical trials. Yet sponsors have historically had limited published evidence to help determine whether observed discordance falls within expected ranges or may warrant closer evaluation.

This publication, led by Gregory Goldmacher, MD, PhD, now Chief Scientific & Medical Officer at Perceptive Imaging, analyzed site-central discordance across a large oncology clinical trial dataset to establish empirical benchmarks across multiple solid tumor types. The findings provide a more evidence-based framework for interpreting observed variability and supporting informed discordance monitoring during trial conduct.

Why Read This Publication?

  • Establish an evidence-based frame of reference for interpreting site-central discordance
  • Understand how expected discordance varies across different solid tumor types
  • Support more informed discordance monitoring during oncology trial conduct
